Glycolipids anchor laminin

چکیده
Boundary proteomics pecialized regions of chromatin flank the edges of transcriptionally silent DNA, separating it from euchromatin. For example, such boundary elements insulate the silent mating type loci ( HMR and HML ) in yeast. The molecular nature of these boundaries has remained obscure. Now, Tackett et al. (page 35) have identified protein complexes that are important for maintaining such boundaries. The team tagged numerous chromatin-associated proteins with protein A and then used a modified form of ChIP to purify protein complexes together with their cognate DNA and nucleosomes. They uncovered several overlapping protein complexes that contained Dpb4, including chromatinremodeling and DNA polymerase holoenzyme complexes. The histones that copurified with these complexes had a unique pattern of acetylation and methylation, intermediate between the patterns seen in active and inactive chromatin. When the team amplified the DNA that purified with the Dpb4 complexes, they found known boundary regions, such as HMR and HML , as well as numerous sequences localized to the ends of chromosomes, and a large number of previously unstudied loci. Significantly, disruption of the protein complexes perturbed boundary function, as detected by the expression of reporter genes inserted in and near the HMR locus. Because the polymerase complex associates with the regions in a cell cycle–dependent manner, Tackett et al. are currently working to test whether it helps conserve the epigenetic signals during replication. They are also investigating the roles of the boundary complexes at the other loci that they identified. S

Laminin-dependent and laminin-independent adhesion of human melanoma cells to sulfatides.
Sulfatides (galactosylceramide-I3-sulfate) but not neutral glycolipids or gangliosides adsorbed on plastic promote adhesion of the human melanoma cell line G361. Direct adhesion of G361 cells requires densities of sulfatide greater than 1 pmol/mm2. In the presence of laminin, however, specific adhesion of G361 cells to sulfatide or seminolipid (galactosylalkylacyl-glycerol-I3-sulfate) but not t...

متن کاملRole of the Cell Wall Phenolic Glycolipid-1 in the Peripheral Nerve Predilection of Mycobacterium leprae
The cell wall of pathogenic mycobacteria is abundant with complex glycolipids whose roles in disease pathogenesis are mostly unknown. Here, we provide evidence for the involvement of the specific trisaccharide unit of the phenolic glycolipid-1 (PGL-1) of Mycobacterium leprae in determining the bacterial predilection to the peripheral nerve. PGL-1 binds specifically to the native laminin-2 in th...
متن کاملLaminins in basement membrane assembly
The heterotrimeric laminins are a defining component of all basement membranes and self-assemble into a cell-associated network. The three short arms of the cross-shaped laminin molecule form the network nodes, with a strict requirement for one α, one β and one γ arm. The globular domain at the end of the long arm binds to cellular receptors, including integrins, α-dystroglycan, heparan sulfate...
متن کاملAn antiglycolipid antibody inhibits Madin-Darby canine kidney cell adhesion to laminin and interferes with basolateral polarization and tight junction formation
Epithelial cells polarize not only in response to cell-cell contacts, but also to contacts with a substratum composed of extracellular matrix molecules. To probe the role of specific matrix constituents in epithelial cell polarization, we investigated the effects of an adhesion-blocking mAb, 12B12, on initial polarization of MDCK cells. The 12B12 antibody, raised against whole MDCK cells, block...
